Semaglutide

Also known as Ozempic, Wegovy

Long-acting GLP-1 receptor agonist.

Medical status: Approved for type 2 diabetes and chronic weight management.

Also commonly used for (off-label)

  • General appetite control and weight loss
  • Reducing food noise and cravings

Off-label and research uses are what people report, not approved indications. Talk to a clinician before using any of them.

How it's taken

Availability varies by supplier — injectable is the most commonly researched form.

Documented dose
250–2500 mcg
Typical frequency
Once weekly, same day each week — hold each dose at least 4 weeks before increasing
Half-life
~7 days
Common vial sizes
5 mg, 10 mg
Regulatory status
FDA-approved prescription medicine.

Dose on a U-100 syringe (250–2500 mcg)

VialBAC waterDraw
5 mg1 mL5.0–50 units
10 mg1.5 mL3.8–38 units
10 mg2 mL5.0–50 units

On a U-100 syringe, 100 units = 1 mL, and the syringe holds 100 units total. Most people keep a single subcutaneous injection under about 50 units — bigger volumes are usually mixed stronger (less water) or split into two shots. Only mixes that fit in one syringe are shown. Always double-check with the reconstitution calculator.

What it does

A long-acting copy of GLP-1, a gut hormone released after eating. It slows stomach emptying and turns down appetite signalling in the brain, so you feel full sooner and stay full longer. Approved as Ozempic and Wegovy. What people look into it for: • Meaningful, sustained weight loss (often 10-15% of body weight in trials) • Quieting constant food thoughts and cravings • Type 2 diabetes and blood-sugar control • Cardiovascular risk reduction shown in large trials Doses are increased slowly because nausea, constipation and reflux are common early on. Losing muscle alongside fat is a real risk without protein and resistance training.

Mechanism

GLP-1 receptor agonist slowing gastric emptying and increasing satiety and insulin secretion.

Storage — lyophilisedwhat's this?

Refrigerate 2-8C, protect from light.

Storage — reconstitutedwhat's this?

Refrigerate 2-8C, use within ~28-56 days per label.

Reconstitution noteswhat's this?

Common mix: 5 mg vial + 1 ml bacteriostatic water = 50 mcg per unit, so 0.25 mg is 5 units, 1 mg is 20 units and 2.5 mg is 50 units. A 10 mg vial + 2 ml gives the same draw.

Stacks well with

Popular combinations reported in the community and research literature — not a recommendation. Adding peptides multiplies side-effect risk; check with a clinician.

Commonly reported side effects

  • Nausea
  • Constipation
  • Vomiting
  • Fatigue
  • Gallbladder issues

Contraindications noted

  • Personal or family history of medullary thyroid carcinoma
  • MEN2
  • Pregnancy
  • History of pancreatitis

Research notes

Titrate slowly; well documented in large randomised trials.
