fat loss

Retatrutide

Also known as LY3437943, Triple-G

Investigational triple GIP/GLP-1/glucagon receptor agonist.

Medical status: Investigational — not approved anywhere yet.

Also commonly used for (off-label)

  • Research into next-generation weight loss
  • Fat loss where GLP-1s have plateaued

Off-label and research uses are what people report, not approved indications. Talk to a clinician before using any of them.

How it's taken (route)

Documented dose
2–12 mg
Typical frequency
Once weekly, same day each week — hold each dose at least 4 weeks before increasing
Half-life
~6 days
Common vial sizes
10 mg, 30 mg, 60 mg
Regulatory status
Investigational; not approved.

Dose on a U-100 syringe (2–12 mg)

VialBAC waterDraw
30 mg1 mL6.7–40 units
60 mg2 mL6.7–40 units
60 mg2.5 mL8.3–50 units

On a U-100 syringe, 100 units = 1 mL, and the syringe holds 100 units total. Most people keep a single subcutaneous injection under about 50 units — bigger volumes are usually mixed stronger (less water) or split into two shots. Only mixes that fit in one syringe are shown. Always double-check with the reconstitution calculator.

What it does

An investigational "triple agonist" that hits three metabolic receptors at once — GLP-1, GIP and glucagon. The glucagon arm also raises energy burn, not just lowers appetite. What people look into it for: • The largest average weight loss reported in this drug class so far in trials • Strong appetite suppression and reduced food cravings • Blood-sugar control and reduced liver fat It is still in clinical trials and is not approved anywhere, so long-term safety is not established.

Mechanism

Triple agonism adds glucagon-receptor-driven energy expenditure to incretin effects.

Storage — lyophilisedwhat's this?

Refrigerate 2-8C, protect from light.

Storage — reconstitutedwhat's this?

Refrigerate 2-8C, use within ~28 days.

Reconstitution noteswhat's this?

Common mix: 30 mg vial + 2.5 ml bacteriostatic water = 120 mcg per unit, so 2 mg is about 16 units, 6 mg is 50 units and 12 mg is 100 units. Typical starting dose is 2 mg per week (4 mg if you have already stepped up on another GLP-1).

Stacks well with

Popular combinations reported in the community and research literature — not a recommendation. Adding peptides multiplies side-effect risk; check with a clinician.

Commonly reported side effects

  • Nausea
  • Vomiting
  • Increased heart rate
  • Diarrhoea

Contraindications noted

  • Pregnancy
  • Thyroid C-cell tumour history
  • Uncontrolled cardiovascular disease

Research notes

Still in clinical trials; no approved human product exists.
